Should I wait a few minutes to reboot, after a Definition Update...?

It isn't. The IKARUS engine is rebuilding it's signature database which includes decrypting and unpacking the current t3sigs.vdb, applying the differential updates obtained from the online update as well as packing and encrypting the newly generated database and saving it to disk.

Q2?- If so, and the update is just a few thousand signatures, and Not a Major... Update (I monitor how long the toolbar icon spins...)-
How long should I wait before a reboot?

Q3?- How long should I wait for a Major update?

It doesn't matter as long as you don't reboot during the update.

Q3?- How long should I wait for a Major update?

Same. As soon as the little info box telling you that an update was applied shows up, you can reboot.
